Plot:

The film opens by introducing Morton Schmidt and Greg Jenko in their respective high school years in 2005. Schmidt is depicted as an awkward, braces-wearing nerd, desperately trying to get a date for prom, only to be rejected and mocked. Jenko, on the other hand, is the popular, athletic jock who bullies Schmidt and enjoys a privileged social status. Seven years later, they both attend the police academy. Despite their contrasting strengths – Jenko excels in physical tasks but struggles with academics, while Schmidt is intellectually sharp but physically inept – they surprisingly become friends, forming an unlikely bond that helps them both graduate. Their initial foray into police work as bicycle patrol officers proves disastrous when they fail to properly read a suspect his Miranda rights during a pursuit, leading to the suspect's release and their suspension from active duty.

As a consequence of their incompetence, they are reassigned to a resurrected undercover program located at 21 Jump Street, operating out of a dilapidated Korean church. Here, they meet their superior, the stern and perpetually angry Captain Dickson, who explains their mission: to infiltrate Sagan High School and identify the supplier of a new synthetic drug called HFS (Holy F***ing Sh**). Dickson warns them that high school has changed since their time, emphasizing the importance of adapting to modern teenage culture. However, a crucial administrative error leads to Jenko and Schmidt swapping identities and schedules. However, the subsequent morning brings renewed arguments, particularly when Schmidt's actions begin to compromise their mission and Jenko feels sidelined.

As the investigation progresses, they discover that Eric is a mid-level dealer and the HFS is being manufactured and distributed on a larger scale. Schmidt, increasingly absorbed in his popular persona and his relationship with Molly, alienates Jenko. Jenko, meanwhile, uses his new connections with the "nerds" to gather intelligence, discovering that the HFS is being produced using chemicals stolen from the school's chemistry lab. This information, combined with Schmidt's insights from the popular crowd, helps them piece together more of the puzzle. Their strained partnership comes to a head during the prom. Jenko, through his nerdy friends, learns that Eric plans a major drug deal at the prom. Schmidt, initially more focused on his date with Molly, eventually realizes the importance of the mission.

At the prom, chaos erupts. Schmidt confronts Eric, who reveals he's merely a distributor and that the real supplier is someone higher up. Mr. Walters, along with his biker gang associates (who were previously involved in the film's opening scene pursuit), intends to distribute a massive shipment of HFS. A frantic chase ensues, involving Jenko, Schmidt, Eric, Molly, and the biker gang. During the chase, Jenko's physical prowess and Schmidt's quick thinking are both put to the test, allowing them to effectively work as a team despite their earlier conflicts.

The chase culminates in a shootout and a chaotic pursuit involving a limousine and a truck. Jenko, utilizing his physical strength, tackles and apprehends several of the bikers, while Schmidt, despite his clumsiness, manages to disarm Mr. Walters. During the confrontation, Mr. Walters reveals his motive: he feels disrespected by the younger generation and wants to corrupt them. Captain Dickson arrives with backup, and Mr. Walters and his associates are arrested. In a moment of genuine heroism, Jenko saves Schmidt from a falling stage prop, solidifying their renewed friendship. Eric, seeing the true nature of his drug-dealing partners, decides to cooperate with the police and becomes an informant, also revealing his desire to attend college and turn his life around.

In the aftermath, Schmidt and Jenko, having proven their worth and matured through their undercover experience, finally earn Captain Dickson's respect.
