Plot:

The story of Partner begins in a coastal town where Sridhar, an earnest and hard-working young man, sees his life upended by financial ruin. Having invested his savings and borrowed substantial sums to operate a prawn farm, an untimely ecological disaster decimates his aquaculture ponds overnight, sinking him into crippling debt. Faced with ruthless moneylenders demanding immediate repayment and desperate to secure a peaceful future for his dependent sister Amudha, Sridhar sees no legitimate avenue to generate fast cash in his hometown. He decides to relocate to Chennai to seek shelter with his childhood buddy, Kalyanaraman, widely known simply as Kalyan. Kalyan lives as a petty criminal and con artist, surviving on stolen goods, quick-buck hustles, and questionable street ventures alongside eccentric associates. Although Sridhar has spent his life striving to remain law-abiding, Kalyan persuades him that a few covert assignments are his only realistic means of clearing his overwhelming liabilities.

Sridhar and Kalyan find themselves pulled into a high-stakes corporate espionage job arranged by John Vijay, a disgruntled former protege of an eccentric elderly research scientist named Prathap. Prathap has spent years holed up in a private suburban laboratory perfecting a miraculous biological compound capable of altering cellular genetics and modifying human body structures. John Vijay, eyeing multimillion-dollar industrial payoffs from crooked politicians and overseas brokers, contracts Kalyan’s crew to infiltrate Prathap’s fortified lab and steal the proprietary prototype serum. Reluctantly agreeing to the heist for a lucrative cut of the spoils, Sridhar and Kalyan sneak into the darkened research compound. However, the amateur burglars bungle the operation when Kalyan stumbles against high-voltage research apparatus and triggers chemical containment vials, inadvertently getting jabbed by an automated syringe filled with the experimental serum before the pair manage to escape into the night.

The following morning, Sridhar awakens in their bachelor hideout to an unfathomable sight. In place of the portly, disheveled Kalyan lying on the adjoining cot, there stands a glamorous, slender young woman. To Sridhar's utter horror and disbelief, the woman begins speaking in Kalyan’s unmistakable, gravelly cadence, utilizing his signature colloquial slangs and exhibiting his coarse physical habits. The experimental serum had triggered an immediate biological metamorphosis, converting Kalyan's physical anatomy into that of a woman while leaving his crude personality, thoughts, and memories completely unaltered. Sridhar is thrown into frantic hysteria as he tries to process that his longtime male best friend is now physically indistinguishable from a high-society fashion model, while Kalyan struggles with sheer panic upon discovering his feminine physique, vocal pitch, and the comical inconveniences of navigating women's attire and physical grooming.

Complicating matters further is Sridhar’s budding romance with Priya, an independent and kind-hearted young woman he met through a chance encounter in the city. Sridhar’s affection for Priya is genuine, but his heart drops into his stomach when he discovers that Priya is the beloved daughter of none other than Scientist Prathap—the very man whose private research facility he and Kalyan had raided the previous evening. Sridhar is forced to juggle maintaining his image as a respectable suitor in front of Priya and her father while secretly hiding the transformed "Kalyani" inside his apartment. Kalyan, now inhabiting Hansika's body, proves to be an uncontrollable domestic handful. He shamelessly burps, sits cross-legged in public dhabas, flirts awkwardly with terrified neighborhood mechanics, and even crosses paths with his former girlfriend Sengamalam, sparking bizarre jealousies and public squabbles across town.

Realizing that Kalyan cannot spend the rest of his life living as a woman, the duo makes desperate covert inquiries to track down an antidote before John Vijay’s syndicate uncovers their failure. However, word of a gorgeous, mysterious woman wandering with Sridhar spreads through Chennai’s underworld, attracting unwanted attention from bumbling gang enforcers Samadhanam and Annadhanam, as well as the corrupt local minister Agnikunju and rival gangster Vikravandi Vaigundam. The criminals mistakenly assume that the female Kalyan is a high-profile mole or an elite courier carrying the stolen formula, sparking a wave of chaotic, slapstick kidnappings and cross-town vehicle chases. Through it all, Kalyan uses his new female persona to deliver unorthodox street fighting moves, taking enemies off guard with aggressive headbutts and brawling tactics completely at odds with his delicate outward appearance.

The escalating turmoil forces Sridhar to confess the entire sequence of events to Priya. While initially shocked and furious at the deceit, Priya recognizes Sridhar’s fundamental decency and his desperate plight to protect his family. Empathizing with the bizarre predicament, she secretly grants them access to her father's laboratory equipment to synthesize an emergency reversing agent before Inspector Om Saravanan closes in on the heist. The climax converges inside Prathap's sprawling research facility where John Vijay and his heavily armed goons storm the premises to seize the formula by force, taking Prathap and Priya hostage. Sridhar, flanked by the fiery female Kalyan and their allies, launches a counterattack to defend the lab, engaging in a frantic physical melee that wrecks the chemical inventories.

Amidst the explosive scuffle, the villains are systematically neutralized, and John Vijay is subdued just as the police arrive to restore order. Prathap, having pieced together the incredible nature of his successful yet chaotic experiment, quickly preps a counter-agent injection to reverse Kalyan’s cellular shift back to normal. However, during the panicked final moments of the lab altercation, the chemical formula is jostled once again, resulting in an errant, second mutation. Instead of returning to Yogi Babu’s familiar look, Kalyan transforms into an entirely new, bald, and intimidating brute played by Mottai Rajendran. Sridhar manages to clear his debts, secure Priya's lifelong partnership, and earn the scientist’s forgiveness, but he is left shaking his head in amused exasperation as Kalyan, now in his newest bizarre physical avatar, steps into the bustling world to start another chaotic day.
